چکیده

Background We’ve previously shown that radiotherapy (RT) converts poorly immunogenic tumors resistant to antibodies (mAbs) against the immune checkpoint receptor CTLA-4 into susceptible ones (Demaria et al., 2005; Dewan et al., 2009). Programmed death-1 (PD-1) is another immune checkpoint receptor upregulated on T-cells shortly after activation and expressed at high levels on exhausted Tcells. Anti-PD-1 mAbs have shown marked clinical activity in some cancer patients, but the majority does not respond. Here we tested the hypothesis that RT can sensitize poorly immunogenic tumors to anti-PD-1.

Beta-adrenoceptor-mediated responsiveness of human internal mammary artery

The internal mammary artery (IMA) is currently the preferred conduit for myocardial revascularization. However, pre-operative vasospasm and a hypoperfusion state during maximal exercise may limit its use as a bypass graft. The mechanism of spasm has not been clearly defined. Since β-adrenoceptor activation plays a major role in vasorelaxation, the present study was carried out to investigate th...

SHORT TERM VARIATION OF ATROPINE BLOCKADE IN THE TRACHEOBRONCHIAL TREE OF ASTHMATIC SUBJECTS

In asthmatic subjects there is a pronounced diurnal variation in bronchial responsiveness. If this phenomenon is due to variation in factors that control drug delivery, then it should be paralleled by a similar variation in competitive antagonist blockade. In order to study this possibility, we performed the methacholine challenge test and after 45 minutes, administered atropine by inhalat...

PD-1 Restrains Radiotherapy-Induced Abscopal Effect.

We investigated the influence of PD-1 expression on the systemic antitumor response (abscopal effect) induced by stereotactic ablative radiotherapy (SABR) in preclinical melanoma and renal cell carcinoma models. We compared the SABR-induced antitumor response in PD-1-expressing wild-type (WT) and PD-1-deficient knockout (KO) mice and found that PD-1 expression compromises the survival of tumor-...

PD-1/PD-L1 blockades in non-small-cell lung cancer therapy

Lung cancer is the leading cause of cancer death in males and the second leading cause of death in females worldwide. Non-small-cell lung cancer (NSCLC) is the main pathological type of lung cancer, and most newly diagnosed NSCLC patients cannot undergo surgery because the disease is already locally advanced or metastatic. Despite chemoradiotherapy and targeted therapy improving clinical outcom...

Local and systemic modulation of the PD-L1 pathway is a novel mechanism of Enzalutamide resistance in castration-resistant prostate cancer

The treatment effects of the anti-androgen Enzalutamide (ENZ) in patients with castration resistant prostate cancer (CRPC) are short lived. Immunotherapy may improve patient survival, however how efficacious these treatments are for CRPC, particularly those that inhibit T cell checkpoint molecules, remains questionable. Indeed, the lack of PD-L1 expression on CRPC tumors has made rationalizing ...
